Targeting the shikimate pathway in the malaria parasite Plasmodium falciparum.
The shikimate pathway presents an attractive target for malaria chemotherapy. Three shikimic acid analogs exhibited different effects on Plasmodium falciparum growth. (6R)-6-Fluoro-shikimate and (6S)-6-fluoro-shikimate inhibited growth (50% inhibitory concentrations, 1.5 x 10(-5) and 2.7 x 10(-4) M, respectively), whereas 2-fluoro-shikimate had no effect. para-Aminobenzoic acid abrogated the in...
متن کاملFate of haem iron in the malaria parasite Plasmodium falciparum.
Chemical analysis has shown that Plasmodium falciparum trophozoites contain 61+/-2% of the iron within parasitized erythrocytes, of which 92+/-6% is located within the food vacuole. Of this, 88+/-9% is in the form of haemozoin. (57)Fe-Mössbauer spectroscopy shows that haemozoin is the only detectable iron species in trophozoites. Electron spectroscopic imaging confirms this conclusion.
متن کاملTranscriptional variation in the malaria parasite Plasmodium falciparum.
Malaria genetic variation has been extensively characterized, but the level of epigenetic plasticity remains largely unexplored. Here we provide a comprehensive characterization of transcriptional variation in the most lethal malaria parasite, Plasmodium falciparum, based on highly accurate transcriptional analysis of isogenic parasite lines grown under homogeneous conditions. This analysis rev...
متن کاملH2O2 dynamics in the malaria parasite Plasmodium falciparum
Hydrogen peroxide is an important antimicrobial agent but is also crucially involved in redox signaling and pathogen-host cell interactions. As a basis for systematically investigating intracellular H2O2 dynamics and regulation in living malaria parasites, we established the genetically encoded fluorescent H2O2 sensors roGFP2-Orp1 and HyPer-3 in Plasmodium falciparum. Both ratiometric redox pro...
متن کاملpH Regulation in the Intracellular Malaria Parasite, Plasmodium falciparum
The mechanism by which the intra-erythrocytic form of the human malaria parasite, Plasmodium falciparum, extrudes H ions and thereby regulates its cytosolic pH (pHi), was investigated using saponin-permeabilized parasitized erythrocytes. The parasite was able both to maintain its resting pHi and to recover from an imposed intracellular acidification in the absence of extracellular Na, thus ruli...
